Choi Dong Yi adalah seorang perempuan dari golongan Cheonmin, kasta rendahan dalam struktur masyarakat Joseon pada masanya. Ayahnya seorang koroner/ ahli forensik yang dibunuh karena dituduh menjadi ketua organisasi terlarang Geumgae yang pada masa itu dianggap sebagai organisasi makar dan membahayakan kekuasaan. Karena sebuah konspirasi politik akhirnya ayah dan kakak Dong Yi meninggal dunia dan ia menjadi gadis belia yang hidup sebatang kara melalui kerasnya hidup. Namun seorang Gisaeng ( wanita yang bekerja di rumah/ timpat hiburan ) cantik bernama Seol Hee telah menyelamatkan dan memeliharanya dengan baik sampai suatu saat Dong Yi berhasil memasuki istana sebagai pelayan di departemen musik.

ki-ka: Raja Sukjong, Dong Yi, Jang Hee Bin, Cha Cheon So

Singkat cerita tanpa sengaja Dong Yi bertemu dengan raja Sukjong yang sedang menyamar untuk menyelidiki sebuah kasus yang dicurigai sebagai usaha konspirasi untuk menyingkirkan calon selir Jang Hee Bin/ Jang Ok Jung. Atas kecerdasan Dong Yi, Jang Hee Bin dapat terlepas dari masalah dan ketika ia menjadi selir, Hee Bin merekomendasikan Dong Yi untuk memasuki Biro Penyelidik Istana. Bukan tanpa perjuangan Dong Yi bisa memasuki departemen ini, sebuah konsprasi politik (lagi-lagi) membuatnya nyaris ditendang dari biro penyelidik ini. Namun karena kecerdasan dan kekerasan hatinya, dong Yi dapat bertahan.

Sayangnya kemudian hubungan Dong Yi dan Hee Bin tidak berjalan mulus, ketamakan Hee Bin dan keluarganya yang dimotori oleh Jang Hae Jee, kakak Hee Bin, membuat Dong Yi menjadi pihak yang berseberangan dengan Hee Bin. Hee Bin sangat berambisi menjadi permaisuri dan berusaha dengan segala cara yang penuh kelicikan menyingkirkan ratu Inhyeon. Ratu Inhyeon sangat menyayangi Dong Yi dan menjadi pelindungnya, bahkan kelak dialah yang mendorong Dong Yi untuk menikah dengan Raja Sukjong dan memintanya menjadi permaisuri sabagai pengganti dirinya saat menjelang kematiannya ( walaupun Dong Yi akhirnya menolak ).

Kemarahan Hee Bin semakin memuncak saat menemukan fakta bahwa sang Raja tidak lagi mencintainya seperti dulu. Raja Sukjong telah berpindah hati pada wanita muda yang cerdas dari biro penyelidik yang dikenalnya tanpa wanita itu tau siapa dirinya. Dong Yi tidak pernah mengetahui bahwa selama ini pejabat yang selalu bersamanya adalah raja Sukjong.

Intrik-intrik politik terjadi, fitnah dan konspirasi mewarnai istana dan begitu banyak korban berjatuhan. Untungnya Dong Yi mempunyai pelindung yang sangat loyal. Mereka adalah Seo Yong Gi ( pejabat kepolisian, mantan teman dekat ayahnya ), Cha Ceon Soo ( seseorang dari masa kecil Dong Yi, mantan anggota Geumgae ) yang juga mencintai Dong Yi, Sim Yun Taek (seorang sarjana cheonmin cerdas yang dibuang ), Ratu Inhyeon dan yang mendukung dari balik layar, Raja Sukjong. Walaupun demikian Dong Yi sempat mengalami masa-masa sulit setelah kematian putra pertamanya dan meninggalkan istana selama 6 tahun. Dalam pengasingannya ia melahirkan putra kedua, Kyeong Jong, yang kelak bergelar Raja Jeongjo, menggantikan kakak tirinya yang meninggal di usia muda.

Choi Sukbin

Di akhir cerita kejahatan akan terkalahkan oleh kebenaran. Setelah hampir sepuluh tahun Hee Bin mengalami kejayaan dengan cara yang licik akhirnya kejayaan yang dimilikinya tumbang. Ia dihukum mati dengan cara minum racun serta keluarga dan kroninya dibuang ke Pulau Jeju untuk dijadikan budak selama-lamanya. Dong Yi menjadi selir tingkat satu bergelar Choi Sukbin, impian ratu Inhyeon untuk menempatkan Dong Yi sebagai penggantinya tidak terwujud karena latar belakan Dong Yi dari kalangan Cheonmin, partai kelas bawah.

Apapun akhir ceritanya yang jelas Dong Yi akhirnya menjadi ibu raja dan hidup bahagia dengan raja Sukjong setelah putranya mengambil alih tahta. Dan yang jelas drama ini benar-benar luar biasa, mengambil setting sejarah Korea yang diwarnai intrik-intrik politik serta konspirasi yang mematikan.
